PHILADELPHIA, Dec. 21, 2022 /PRNewswire/ -- Researchers from Children's Hospital of Philadelphia (CHOP) and New Jersey Institute of Technology (NJIT) developed new software that integrates a variety of information from a single cell, allowing researchers to see how one change in a cell can lead to several others and providing important clues for pinpointing the exact causes of genetic-based diseases.

Key Points:
  • Single-cell sequencing allows researchers to look at specific aspects of a cell to determine how it interacts with its microenvironment.
  • At the single- cell level, researchers can study gene expression as well as messenger RNA, proteins and even organelles within the cells in much greater detail and resolution than before.
  • The software, referred to as single-cell multimodal deep clustering (scMDC), uses machine learning to analyze data about different characteristics of a single cell.
  • The researchers conducted extensive simulation and real-data experiments and found that scMDC outperformed existing single cell single-modal and multimodal clustering methods on single-cell multimodal data sets.

CHOP-led Study Shows that Antibiotic-Resistant Microbes in the Gut Make C. difficile More Infectious

Retrieved on:
Wednesday, November 16, 2022

PHILADELPHIA, Nov. 16, 2022 /PRNewswire/ -- Clostridioides difficile, often referred to as C. difficile or C. diff, is a bacterium that causes severe intestinal illness and, as its name suggests, can be difficult to study and treat. Approximately 1 in 6 patients infected with C. difficile will be reinfected within two months. Yet scientists have not figured out why C. difficile infection is more difficult to treat in some patients versus others. The human gut is filled with trillions of microbes, and these microbes influence the virulence of various pathogens, but until now, scientists had little understanding of how C. difficile cooperates with the rich collection of microorganisms in the gastrointestinal tract.

Key Points:
  • Yet scientists have not figured out why C. difficile infection is more difficult to treat in some patients versus others.
  • Prior studies have shown that adults infected with C. difficile also have high levels of Enterococcus in their gut and that vancomycin-resistant Enterococcus (VRE) frequently co-infects patients with C. difficile.
  • They also observed a positive correlation between C. difficile burdens and ornithine, supporting a key role for this amino acid in C. difficile infection.

PHILADELPHIA, Nov. 15, 2022 /PRNewswire/ -- Financial stress due to the COVID-19 pandemic took a distinct toll on adolescent mental health and contributed to depressive symptoms, according to a new study by researchers at Children's Hospital of Philadelphia (CHOP). The study, published today in The Lancet Regional Health – Americas, found the effect was most pronounced in low-income adolescents but also affected all income groups who experienced financial strain due to loss of income.

Key Points:
  • However, despite an ongoing global youth mental health crisis, there previously was little data on the impact of financial stress on adolescent mental health.
  • The researchers investigated the specific association of financial strain with adolescent mental health between May 2020 and May 2021.
  • "Future studies should look at targeting youth's financial stress and family conflict to ease the mental health impact of difficult financial circumstances."

CHOP Researchers Find COVID-19 Vaccination Leads to Higher Antibody Levels than Natural Infection in Both Pregnant People and their Babies

Retrieved on:
Wednesday, November 9, 2022

PHILADELPHIA, Nov. 9, 2022 /PRNewswire/ --Pregnant people who received one of the mRNA COVID-19 vaccines had 10-fold higher antibody concentrations than those who were naturally infected with SARS-CoV-2, a finding that was also observed in their babies, according to a new study by researchers at Children's Hospital of Philadelphia (CHOP) and the University of Pennsylvania. The study, published today in JAMA Network Open, also found that vaccine timing played an important role in maximizing the transfer of antibodies, with antibodies detected as early as 15 days after the first vaccine dose and increasing for several weeks after.

Key Points:
  • "Given that pregnancy is a risk factor for severe COVID-19, this study suggests pregnant people should prioritize getting vaccinated to protect themselves and their babies."
  • In collaboration with CHOP researchers, his laboratory evaluated cord blood serum from 585 pregnant people who had detectable SARS-CoV-2 antibodies.
  • They found that antibody levels among vaccine recipients were approximately 10-times higher than in people who had been naturally infected.
  • Of those newborns with detectable antibodies, the researchers found that levels were 10-times higher in the vaccinated group than in the naturally infected group.
